چکیده

The INO80 and SWR1 complexes belong to the INO80 subfamily of ATP-dependent chromatin-remodeling complexes. These complexes interact with nucleosomes and remodel chromatin by either sliding nucleosomes along the DNA or exchanging histones within nucleosomes. In doing so, the INO80 and SWR1 complexes promote recruitment of regulatory factors to DNA. Distinct from other subfamilies of chromatin-remodeling complexes, INO80 and SWR1 contain core ATPases with a unique split ATPase domain, as well as Rvb proteins, which share limited homology to bacterial RuvB, the Holliday junction DNA helicase. They bind to histone H2A variants (H2AX and H2A.Z) and have been implicated in the alteration of chromatin structure during transcription, replication, and DNA repair. This SnapShot depicts these two remodeling complexes, their mechanism of action, and their biological functions.

Distinct roles for SWR1 and INO80 chromatin remodeling complexes at chromosomal double-strand breaks.

INO80 and SWR1 are two closely related ATP-dependent chromatin remodeling complexes that share several subunits. Ino80 was reported to be recruited to the HO endonuclease-induced double-strand break (DSB) at the budding yeast mating-type locus, MAT. We find Swr1 similarly recruited in a manner dependent on the phosphorylation of H2A (gammaH2AX). This is not unique to cleavage at MAT; both Swr1 ...

Interplay between Ino80 and Swr1 chromatin remodeling enzymes regulates cell cycle checkpoint adaptation in response to DNA damage.

Ino80 and Swr1 are ATP-dependent chromatin remodeling enzymes that have been implicated in DNA repair. Here we show that Ino80 is required for cell cycle checkpoint adaptation in response to a persistent DNA double-strand break (DSB). The failure of cells lacking Ino80 to escape checkpoint arrest correlates with an inability to maintain high levels of histone H2AX phosphorylation and an increas...

Unique subunits Swp82, Taf14, Snf6, Snf11 Rsc3,5,7,9, 10,30, Htl1, Ldb7, and Rtt102 INO80 regulates transcription and is involved in DNA repair and cellcycle checkpoint adaptation. It has ATP-dependent nucleosome mobilization activity in vitro. SWR1 regulates transcription and is also involved in deposition of histone H2AZ, chromosome stability, and cell-cycle checkpoint adaptation. In vitro it...

SWR-C and INO80 Chromatin Remodelers Recognize Nucleosome-free Regions Near +1 Nucleosomes

SWR-C/SWR1 and INO80 are multisubunit complexes that catalyze the deposition and removal, respectively, of histone variant H2A.Z from the first nucleosome at the start of genes. How they target and engage these +1 nucleosomes is unclear. Using ChIP-exo, we identified the subnucleosomal placement of 20 of their subunits across the yeast genome. The Swc2 subunit of SWR-C bound a narrowly defined ...

The INO80 chromatin remodeler sustains metabolic stability by promoting TOR signaling and regulating histone acetylation

Chromatin remodeling complexes are essential for gene expression programs that coordinate cell function with metabolic status. However, how these remodelers are integrated in metabolic stability pathways is not well known. Here, we report an expansive genetic screen with chromatin remodelers and metabolic regulators in Saccharomyces cerevisiae. We found that, unlike the SWR1 remodeler, the INO8...
